چکیده

Bacterial specialized metabolites are of immense importance because their medicinal, industrial, and agricultural applications. Streptomyces clavuligerus is a known producer such compounds; however, much its metabolic potential remains unknown, as many associated biosynthetic gene clusters silent or expressed at low levels. The overexpression ribosome recycling factor (frr) engineering (induced rpsL mutations) in other spp. has been reported to increase the production metabolites. Therefore, we used an strategy combination with untargeted metabolomics, molecular networking, silico analysis annotate 28 current study, which have not previously S. clavuligerus. Many newly described commonly found plants, further alluding ability produce compounds under specific conditions. In addition, manipulation frr led different metabolite profiles most cases. Known putative observed also discussed. This work suggests that traditional strain recently developed metabolomics technologies together can provide rapid cost-effective strategies speed up discovery novel natural products.

Construction of Recombinant Vectors Containing Clavulanic Acid Antibiotic Regulatory Gene, claR, Isolated of Streptomyces clavuligerus Strains

The claR of Streptomyces clavuligerus in the clavulanic acid gene cluster encodes a transcriptional regulator that controls clavulanic acid biosynthesis. The main goal of this study was isolation and molecular detection of the claR gene and its cloning in the Streptomyces specific vector (pMA:: hyg). By cinsideration of the claR gene’s start codon, the specific primers were designed. After geno...

Engineered Streptomyces platensis strains that overproduce antibiotics platensimycin and platencin.

Platensimycin, which is isolated from Streptomyces platensis MA7327, and platencin, which is isolated from S. platensis MA7339, are two recently discovered natural products that serve as important antibiotic leads. Here we report on the identification of S. platensis MA7327 as a dual producer of both platensimycin and platencin. A PCR-based approach was used to locate and clone the locus involv...
